Description: Sfi I is a Restriction Endonuclease purified from an Escherichia coli strain that carries the Sfi I gene from Streptomyces fimbriatus. Sfi I activity is often affected by CG methylase. When the sequence containing the recognition site is as follows, the enzyme can be affected by dcm methylase and would not digest DNA substrates:

NZYTech provides Sfi I with a specific buffer, NZYBuffer A. NZYTech’s Sfi I is an enzyme that acts at 50 °C. For double digestions please use NZYBuffer U (not provided, MB110).

Reagents supplied with the enzyme: 10× NZYBuffer A (500 mM Tris-HCl, pH 7.5, 1 M NaCl, 100 mM MgCl2, 10 mM DTT).
Storage buffer: 10 mM Tris-HCl, pH 7.5, 200 mM NaCl, 0.1 mM EDTA, 7 mM 2-mercaptoethanol, 200 µg/mL (w/v) BSA, 50% (v/v) glycerol.
