Description: NZY RNase H is a recombinant endoribonuclease purified from anEscherichia coli strain that carries the cloned RNase H gene (rnh). The enzyme specifically hydrolyses the phosphodiester bonds of RNA which is hybridized to DNA. RNase H does not degrade single and double-stranded DNA or unhybridized RNA.  It is a key enzyme in the removal of mRNA after first-strand cDNA synthesis. In addition, RNase H is useful for the removal of poly(A) tails on mRNAs after hybridizarion with oligo (dT), and for oligodeoxyribonucleotide-directed site-specific cleavage of RNA.

Shipping and Storage conditions: Shipped refrigerated. NZY RNase H should be stored at -20 °C in a freezer without a defrost cycle. The enzyme will remain stable up to 3 years if stored as specified.

Storage buffer: 20 mM Tris-HCl, pH 7.9, 100 mM KCl, 10 mM MgCl₂, 0.1 mM EDTA, 0.1 mM DTT and 50% (v/v) glycerol.

Concentration: 5 U/µl

Reaction buffer (10×): 200 mM Tris-HCl, pH 8.4, 500 mM KCl, 50 mM MgCl₂, 200 mM DTT.
