Description: NZYLong 2× Green Master Mix is a premixed ready to use solution containing an optimized blend of NZYTaq DNA polymerase and NZYProof DNA polymerase designed to amplify target DNA sequences ranging from 1 to more than 15 kb in size. The proofreading activity of NZYProof DNA polymerase, enables NZYLong DNA polymerase to display higher fidelity than Taq DNA Polymerase alone. The master mix contains dNTPs and reaction buffer at optimal concentrations for efficient amplification of DNA templates by PCR. MgCl₂ final concentration is 2.5 mM, allowing the implementation of a comprehensive range of PCR protocols. In addition, reactions assembled with NZYLong 2× Green Master Mix maybe directly loaded onto agarose gels. NZYLong 2× Green Master Mix contains two dyes (blue and yellow) that allow monitoring the progress of electrophoresis. NZYLong 2× Green Master Mix is not suitable when direct fluorescent or absorbance readings are required without prior purification of the amplified DNA from PCR. Resulting polymerase chain reaction (PCR) products have an A overhang and are suitable for cloning with NZYTech´s TA PCR cloning kits (MB053 or MB137).

 

Storage temperature: NZYLong 2x Green Master mix should be stored at -20 °C, in a constant temperature freezer. Minimize the number of freeze-thaw cycles by storing in working aliquots. The Mix maybe stored at 4 °C for up to 7 weeks.

 

Storage buffer: 20 mM Tris-HCl, pH 7.5, 100 mM NaCl, 0.1 mM EDTA, 1 mM DTT, 50% (v/v) glycerol.
