Description: Supreme NZYTaq 2× Green Master Mix with separate MgCl₂ is a premixed ready to use solution containing a recombinant modified form of Taq DNA polymerase with a hot-start like PCR capacity. The enzyme is inactive at room temperature, avoiding extension of non-specifically annealed primers or primer-dimers and providing higher specificity of DNA amplification. The functional activity of the enzyme is restored during a short 5-minute incubation step at 95 °C. 
In addition, the master mix contains dNTPs and reaction buffer at optimal concentrations for efficient amplification of DNA templates by PCR. NZYTech standard master mix (MB054) contains MgCl₂ at a 2.5 mM concentration. Occasionally some primers and DNA templates require slightly adjusted MgCl₂ concentrations for optimum results. Therefore, MgCl₂ is provided separately in this mix allowing for the MgCl₂ optimization in any PCR system. Reactions assembled with this Master Mix maybe directly loaded onto agarose gels. Supreme NZYTaq 2× Green Master Mix with separate MgCl₂ contains two dyes (blue and yellow) that allow monitoring the progress of electrophoresis. Supreme NZYTaq 2× Green Master Mix with separate MgCl₂ is not suitable when direct fluorescent or absorbance readings are required without prior purification of the amplified DNA from PCR. 
Supreme NZYTaq DNA polymerase is a broad range enzyme suitable for a variety of general PCR applications to amplify target DNA sequences up to 3 kb in size. It enhances many complex PCR reactions by increasing both specificity and yield in a range of DNA concentrations up to 5-10 pg. The activated enzyme maintains the same functionality as TaqDNA polymerase. Supreme NZYTaq DNA polymerase lacks 3´-5´ exonuclease activity. Resulting polymerase chain reaction (PCR) products have an A overhang and are suitable for cloning with NZYTech´s NZYPCR cloning kit (MB01301 or MB01302).

Storage temperature: Supreme NZYTaq 2× Green Master Mix and MgCl₂ should be stored at -20 °C, in a constant temperature freezer. Minimize the number of freeze-thaw cycles by storing in working aliquots. The Mix maybe stored at 4 °C for up to 7 weeks.
